Description
Melanotan 2 Acetate is a conformationally constrained cyclic lactam heptapeptide (Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH₂) designed as a superpotent analog of native α-MSH. It functions as a non-selective agonist at melanocortin receptors MC1R–MC5R. Binding to MC1R on melanocytes elevates intracellular cAMP, activates the MITF pathway, and upregulates tyrosinase, driving eumelanin synthesis and skin pigmentation. Concurrent activation of central MC3R and MC4R receptors modulates hypothalamic circuits involved in sexual motivation, erectile signaling, and energy balance, producing the secondary effects of increased libido and reduced appetite observed in preclinical and limited early-phase human studies.​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​​
