Melanotan 2 Acetate is a conformationally constrained cyclic lactam heptapeptide (Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH₂) designed as a superpotent analog of native α-MSH. It functions as a non-selective agonist at melanocortin receptors MC1R–MC5R. Binding to MC1R on melanocytes elevates intracellular cAMP, activates the MITF pathway, and upregulates tyrosinase, driving eumelanin synthesis and skin pigmentation.
